Results and Discussion. The concentration of the elastase 71proteinase inhibitor complex in synovial fluid ranges from 0 to about 100,000 gg/1 depending on the degree of inflammation and shows an approximately logarithmic distribution. As expected there is a significant correlation between the neutrophils (x) and the elastase inhibitor complex (y) (r = 0.85, log y = 1.02 log x 0.75). In inflammatory joint diseases an increase of the enzyme inhibitor complex is observed (Table 1). No elastase activity is detectable in synovial fluids from noninflammatory joint diseases, but our results show that in some cases of inflammatory joint diseases with a high count of granulocytes enzyme activity can be measured in the synovial fluid. The failure to detect elastase activity in recent studies [2] has been attributed to the fact that the released enzyme is neutralized outside the cell by complex-formation with proteinase inhibitors, mainly ~l-proteinase inhibitor. Elastase and cO-proteinase inhibitor react stoechiometrically and are bound covalently. The activity of the enzyme is completely abolished. However, the inhibition of the elastase activity by 72-macroglobulin only results in a steric hindrance of the proteinase active site. Thus the

activity of such complexes is much lower on protein substrates, but a well defined activity could be demonstrated especially towards low molecular substrates [5]. The proteinase inhibitors of elastase are present in the synovial fluid in a considerable excess, as shown in Table 1. Therefore it seems likely that the determined elastase activity is not referred to free elastase but due to complexes of the proteinase with e2-macroglobulin, which hydrolyze low molecular weight substrates.

In fasting sera, the total cholesterol can be classified into 3 major fractions: the atherogenic low-density lipoprotein cholesterol (LDL-C) and the non-atherogenic very-low-density and highdensity lipoprotein cholesterol (VLDL-C and HDL-C). HDL-C is simple to determine by polyanion precipitation methods, while the differentiation of VLDL-C and LDL-C is still an analytical problem. LDL-C can be calculated from total cholesterol, if, in addition to HDL-C, VLDL-C is known. We have compared 3 different methods for VLDL-C determination (see Table 1):

436 C. Quantitative lipoprotein electrophoresis according to Wietand and Seidel [2]. A nomogram has been developed for the simple evaluation of electropherograms. Table I shows that, at triacylglycerol concentrations above 2 g/l, VLDL-C may be greatly overestimated (and consequently LDL-C underestimated), if the Friedewald method is used. In these 8 cases VLDL-C from ultracentrifugation is "triacylglycerol/7" rather than "triacylglycerol/5" with extreme values ranging from "triacylglycerol/5" to "triacylglycerol/18". In contrast, the lipoprotein electrophoresis yields values which correspond well to the ultracentrifugation data. Comparison of methods B and C in 51 patients showed a significant correlation (r = 0.781, P < 0.001). As a mean, pre-fl-cholesterol from electrophoresis was equivalent to one eighth of total serum tri-
acylglycerol yet with extreme values ranging from triacylglycerol/3 to triacylglycerol/24. From these data, it is concluded that VLDL-C can be determined by ultracentrifugation or lipoprotein electrophoresis, while, at least at triacylglycerol concentrations above 2 g/l, the Friedewald method often yields incorrect results.

1. Einleitung. [mmobilisierte PhenylboronsS,ure wird zur Abtrenhung der Catecholamine im Urin und Plasma benutzt [1 - 3, 6]. Bei einer anschliegenden Bestimmung der Catecholamine mit Hochdruekflfissigkeits-Chromatographie und amperometrischer Detektion werden eine Reihe von St6rsubstanzen nachgewiesen. Aus diesem Grund wurde versucht, eine Abtrennung der Catecholamine mit einem schwach sauren Ionenaustauscher durchzufiihren. 2. Arbeitsweise. Auf das mit Waschpuffer (0,2 mol/l Natriumphosphatpuffer, pH 6,6)/iquilibrierte Bors/iuregel (3,5 x 0,9 cm) wurden in l ml Waschpuffer eine Reihe von Testsubstanzen aufgetragen und ihr Retentionsvolumen gemessen. Eluiert wurden in 7 ml Waschpuffer die S/iuren, Alkohole und AminosS, uren (Tabelle 1 a). Die Elution der Amine erfolgte anschlieBend mit 0,06 mol/1 Salzs/iure (Tabelle lb). Mit dem Ionenaustauscher (Amberlite CG 50 II, 2,5 • 0,9 cm) erfolgte die ,Aquilibrierung, das Auftragen der Testsubstanzen und Waschen der S/iulen in der gleichen Weise. Die Elution der Amine liel3 sich mit 0,8 mol/1 Bors/iure durchffihren. 3. Ergebnisse und Diskussion. Higa [2], der die Abtrennung der Catecholamine auf Bors/iuregel als erster angewendet hat, postulierte in Analogie zur Komplexbildung aliphatischer cis-Diole [5] die Bildung cyclischer Bors/iureester mit den phenolischen Gruppen der Catecholamine gemfiB (1 und 2)

Wfirde dieser Mechanismus zutreffen, so miiBten mittels Borsfiuregel Dihydroxyverbindungen von methylierten oder monohydroxylierten Verbindungen abgetrennt werden. Die Retention der O-methylierten Amine und das Ausbleiben einer Adsorption der Dihydroxyphenyls~iuren macht aber deutlich, dab unter den gegebenen Bedingungen die Ausbildung cyclischer Bors~iureester keine wesentliche RoUe spielt. Die von Roy u.a. [4] bestimmten Komplexbildungskonstanten des Brenzcatechins sind sicher nicht fibertragbar, wenn das angreifende Diol zugleich eine positiv geladene Aminogruppe enthNt. Je kleiner die Dissoziationskonstante des Salzes, um so gr613er muB das Retentionsvolumen des Amins sein. Entsprechend wird Dopamin, dessen Phenylborons/iuresalz wasserlOslich ist, frtih, das Dimethoxyphenylethylamin, dessen Phenylborons/iuresalz schwer wasserl6slich ist, sprit eluiert.
