Kopf- u n d Schwanzanlage waren bei ihnen racist hochgradig miBgebildet. Bei den iiberlebenden K e i m e n war die weitere E n t w i c k l u n g deutlich verlangsamt. Sic zeigten starke Fehlentwicklungen des Kopfteils u n d alle tJbergAnge v o m v611igen Fehlen des I~opfes bis zur Normalform. D a n e b e n fanden sich nicht selten eine Aplasie oder Hypoplasie der Augen, MiBbildungen der Kiemensti~mmchen, V e r k r t h n m u n g e n der L~ngsachse, verkfirzte Schw~nze, hydropische Auftreibungen der Herzbucht. 3. Histologisch zeigten Keime im S e h w a n z k n o s p e n s t a d i u m m e h r oder minder starke Ver~nderungen des Neuroektoderms u n d Mesoderms. Das Neuralrohr fehlte v611ig, oder es war unterentwickelt u n d ungeordnet. Dabei war die ZahI der Neuralzellen u n d der Mitosefiguren wesentlich geringer als im normalen Neuratrohr. I m Mesoderm Ianden sich zahlreiche Kernpyknosen, besonders a u c h bei v611igem Fehlen des Neuralrohrs zwischen Chorda u n d H a u t e k t o d e r m . Bei weiterentwickelten E m b r y o n e n waren die Anlagen yon Hirn, Rtickenmark u n d Augen stark gehemmt. F U k a n n also auch a m T r i t u r u s - K e i m zu MH3bildungen ftihren. Frflher wurde gezeigt, dai3 an T r i t u r u s - K e i m e n durch 5-Fluorouracil (FU) MiBbildungen verschiedener Art hervorgerufen werden k6nnen [1 ~. F U wird im Organismus in 5-Fluorodesoxyuridin (FUDR) umgewandelt. Dieses verhindert dutch H e m m u n g der T h y m i d i n - S y n t h e t a s e die Methylierung yon Desoxyuridylsgmre zu T h y m i n s ~ u r e u n d verursacht dadurch eine H e m m u n g der D N S - S y n t h e s e n [2]. I n dieser Arbeit wurden Keime yon Xenopus laevis bei normaler Entwicklung bzw. u n t e r Einwirkung yon F U zur elektiven Markierung der D N S - S y n t h e s e n m i t Thymidin-~H behandelt. J u n g e Neurulae wurden in 1/2 verdflnnter HoltfreterL 6 s n n g mit F U in 5 0 - - t 0 0 m g / l 6 - - 1 5 h lang gehalten. Thymidin-3H wurde im frahen S c h w a n z k n o s p e n s t a d i u m einmalig 3 h zur Einwirkung gebracht. Dazu wurde der kaudale K6rperpol des Keimes abgetrennt, u m die Isotopen v o n d e r SchnittflXche aus einzubringen. Die Keime wurden in Serien geschnitten u n d n a c h U b e r s c h i c h t u n g der Schnittserien mit S a k u r a - E m u l s i o n in Gel-Form N R - M I oder N R - E 1 nach Exposition yon 3 bzw. 4 Wochen histoautoradiographisch durchgearbeitet. Die Versuche h a t t e n folgende Ergebnisse: I. Bei den Normalkeimen wurden die neuroektodermalen u n d die dorsalen mesodermalen Zellen sehr bevorzugt durch Thymidin-3H markiert. Die Markierung war intensiv u n d g u t auswertbar, die Silberk6rner fanden sich ausschlieglich iiber vielen Kernen der g e n a n n t e n Keimteile. Das Markierungsm u s t e r war ~hnlich wie das an T r i t u r u s - K e i m e n [3J. 2. Die Zahl der markierten Kerne pro gesamter Kernzahl in den verschiedenen Keimteilen (all-Index) war n a c h 6 h Einwirkung yon F U 50--100 rag/1 in den neuroektodermalen Zellen leicht herabgesetzt, noch nicht dagegen im Mesoderm u n d H a u t ektoderm. 3. N a c h 15 h Einwirkung yon F U betrug der 3HIndex bei 50 rag/1 F U 48%, bei 100 mg/1 F U 34% gegeniiber 55% beim Normalkeim. ~ h n l i c h war die R e d u k t i o n im dorsalen Mesoderm u n d im H a u t e k t o d e r m . Es k a m also zur ai1gemeinen H e m m u n g der D N S - S y n t h e s e bei geniigend langer und intensiver Eimvirkung yon FU. Besonders bevorzugt war

diese H e m m u n g an den Neuralzellen des ZNS im Kopfbereich, vor allem n a e h 100 mg/1 FU. 4. Die Silberkornzahl pro Zellkern war in der Hirnanlage der Versuchskeime eindeutig vermindert. Aus den U n t e r s u c h u n g e n schlieBen wir, dab a m Amphibienkeim die D N S - S y n t h e s e dutch F U bei gentigend langer Einwirkung u n d K o n z e n t r a t i o n stark g e h e m m t wird. A Non-Radiative Photochemical Model for Polycyclic Aromatic Hydrocarbon-Induced Carcinogenesis N. P. BuIJ-Ho~ and S. S. SIJNG Centre National de la Recherche Seientifique, Paris, France It is now generally accepted t h a t one of the early events in the carcinogenetic process induced by polyeyclic aromatic hydrocarbons is a reaction of the responsible c o m p o u n d with certain cellular substrates (proteins and/or nucleic acids). B u t the i n t i m a t e m e c h a n i s m of such a reaction remains unknown. E x p e r i m e n t a l and theoretical d a t a suggest, however, t h a t the reaction between carcinogen and cell s u b s t r a t e could be of a photochemical type. Thus, benzo~a]pyrene is known to react with D N A when irradiated with light corresponding to one of its absorption bands [t], and the carcinogenicity of this hydrocarbon is potentiated by light [2]. Further, a positive correlation has been found between carcinogenic activities of polycyclic hydrocarbons and experimental excitation energies on the one h a n d [3], and p h o t o d y n a m i c activity on the other [4, 5]. The difficulty remained to find a source for the energization of t h e postulated " p h o t o c h e m i c a l " reaction in conditions of absence of light. Evidence t h a t chemiluminescent reactions could serve as an excitation energy generator to promote "dark" photochemical reactions was recently supplied by White et al. E6] who found t h a t thermodecomposition of 2-oxaoxetanes generates f r a g m e n t molecules in electronically excited states, and t h a t this excitation energy can serve to promote photoisomerizations instead of undergoing radiative dissipation. Biochemiluminescent reactions in higher organisms are becoming more familiar, following recent investigations [7], and it is known t h a t both the oxidation of polycyclic hydrocarbons [8] and the decomposition of their peroxides [9] can be luminescent; nor can the endogenous formation of 2-oxaoxetanes and c o m p o u n d s with similar properties in the course of normal or pathological metabolism be excluded. Either the postulated photochemical reaction between carcinogen and cell s u b s t r a t e to form a complex could be sufficient to initiate the carcinogenetic process; or the s u b s t r a t e moiety could sustain further alteration b y repeated electronic excitation of the hydrocarbon moiety followed b y transfer of this energy to selective parts of the s u b s t r a t e moiety; this process would be similar to the photo-inactivation of t h e combination Epyridoxal p h o s p h a t e + 6-phosphogluconate dehydrogenase], in which the pyridoxal moiety functions as a "molecular lightning r o d " for the transfer of its photoexcitation energy to the protein nmiety, with ensuing d e n a t u r a tion through t h e oxidation of the histidine residues in the neighbouring active site of the enzyme [10]. The present model lends itself to experimental veri/ication in several ways: in particular, it is to be expected t h a t extensive deuteration, which enhances the life s p a n of triplet states of aromatic hydrocarbons It t l, would also be favonrable to carcinogenic activity, and t h a t simultaneous in situ irradiation with light of appropriate wave-lengths would confer substantial carcinogenieity on those hydrocarbons which of themselves possess only marginal oneogenic properties.
