W h e n w o r k i n g w i t h h o m o g e n a t e s of s e a u r c h i n eggs, therefore, it appears important to be able to check the a m o u n t of c o n t a m i n a t i n g s e a w a t e r . W e h a v e f o u n d t h a t t h e m o s t c o n v e n i e n t w a y t o d o t h i s is t h e e s t i m a t i o n of N a +. I t is k n o w n i n f a c t t h a t t h e N a + c o n t e n t of s e a u r c h i n s a n d s t a r f i s h egg 4 is v e r y small, t h e p r e v a i l i n g i o n b e i n g K + T h e r e f o r e , f r o m t h e e s t i m a t e of N a + in a n y p r e p a r a t i o n , t h e a m o u n t of s e a w a t e r p r e s e n t c a n b e calculated quite accurately. In our practice Na + has been determined by flame-photometry~ O u r t e s t s y s t e m c o n s i s t e d of a p r e p a r a t i o n of r a t l i v e r mitochondria; cytochrome oxidase has been estimated m a n o m e t r i c a l l y as d e s c r i b e d b y MAGGIO a n d MONROY 1 a n d esterified P w a s d e t e r m i n e d e n z y m a t i c a l l y as G - 6 - P b y t h e use of t h e d e h y d r o g e n a s e ~.

S t r u k t u r e n in vielgestaltiger, m e h r o d e r w e n i g e r k o m p a k t e r F o r m , w o b e i w i e d e r u m die B a k t e r i e n - D N S in d e r K e r n v a k u o l e wie a u c h die reine D N S a u s s e r h a l b d e r B a k t e r i e n z e t l e n s e h r Ahnliche K o n f i g u r a t i o n e n z e i g t e n ( F i g u r e n 5 u n d 6). Z u s a m m e n f a s s e n d k 6 n n e n wir sagen, d a s s sich bei gleichert P r A p a r a t i o n s b e d i n g u n g e n reine D N S - K o l l o i d e verschiedenster Herkunft (Forellensperma, Kalbsthymus) v611ig gleich v e r h a l t e n in ihrer A n t w o r t auf die p r ~ p a r a t i v e n Eingriffe wie die D N S in d e r K e r n v a k u o l e d e r B a k terien. Das feinf/~dige Bild der D N S , wie es F i g u r 1, 3 u n d 4 zeigen, e n t s p r i c h t e i n i g e r m a s s e n d e m , was m a n sich u n t e r einer kolloidalen L 6 s u n g vorstellt. I)agegen w i r d d o c h sicherlich d e r A s p e k t in F i g u r 2, a b e r a u c h in F i g u r 5 u n d F i g u r 6 wohl y o n j e d e r m a n n als eine K o a g u l a t i o n angesprochen. E s m u s s sich die B a k t e r i e n - D N S des N u k l e o i d s in e i n e m physikalisch-chemischen Zustand befinden, der nicht welt e n t f e r n t sein k a n n y o n d e m j e n i g e n r e i n e r D N S . O b u n d in
w e l c h e r F o r m an die I)NS g e b u n d e n e H i s t o n e und P r o t a m i n e das R e s u l t a t beeinflussen, wird gegenw/irtig u n t e r s u c h t . Auf d e n O r g a n i s a t i o n s z u s t a n d d e r B a k t e r i e n - l ) N S k a n n bier n i c h t im e i n z e l n e n e i n g e g a n g e n w e r d e n . In d e r M e h r z a h l d e r Fiille e r s c h e i n t d a s feinfXdige D N S - P l a s m a b i i n d e l f 6 r m i g a n g e o r d n e t ~,~4as. Aus diesen B e o b a c h t u n g e n , z u s a m m e n m i t frtiheren und anderen noch nicht mitgeteilten, ergibt sich, dass die O s m i u m f i x i e r u n g noch nicht das D N S - K o l l o i d oder das D N S - P l a s m a ~fixiert~ im Sinne einer Verfestigung, sondern gewissermassen die Weiche stellt ftir die Vorg~inge, die sparer bei der EntwAsserung stattfinden. Es w u r d e y o n KELLENBERGER 16 die F o r d e r u n g aufgestellt, dass das D N S-Kotloid so v o r z u b e h a n d e l n sei, dass bei der D e h y d r a tisierung V e r n e t z u n g e n entstehen. D a d u r c h geliert das Kolloid zuerst, was zur Folge hat, dass die nachfolgende K o a g u l a t i o n , beziehungsweise das A n e i n a n d e r l a g e r n der F a d e n m o l e k f i l e auf ein M i n i m u m beschrAnkt w e r d e n kann. Zus~itzliche E i n z e l h e i t e n dieser A r b e i t e n und nachfolgende Diskussionen der R e s u l t a t e werden an anderer Stelle m i t g e t e i l t werden 1~. S u m m a r y . N o t o n l y h a v e m a n y of t h e p r o b l e m s of i m m u n o f l u o r e s c e n c e been inherited, b u t it has become a p p a r e n t t h a t o t h e r p r o b l e m s exist. Some of the p r o b l e m s e n c o u n t e r e d are presented. I t is hoped t h a t o t h e r p r o b l e m s can be p r e d i c t e d early in t h e d e v e l o p m e n t of t h e t e c h n i q u e , to p r o v i d e a m o r e orderly a n d realistic a p p r o a c h t h a n was e n c o u n t e r e d in immunofluorescence. Methods and Results. F i x a t i o n . U n f i x e d and 10% formalin-fixed chorioallantoic m e m b r a n e and n o r m a l m o u s e liver, kidney, heart, and spleen were t r e a t e d w i t h an i m m u n e Staphylococcus aureus ferritin c o n j u g a t e 1 for 1 h; t h e t r e a t e d m a t e r i a l was t h e n t h o r o u g h l y washed and the unfixed m a t e r i a l fixed in 10% formalin. The specimens were e m b e d d e d in paraffin and 4~-sections stained w i t h G o m o r i ' s iron stain a. As d e m o n s t r a t e d b y t h e iron stain, t h e tissues fixed prior to t r e a t m e n t w i t h t h e i m m u n e conj u g a t e e x h i b i t e d a m a r k e d d i m i n u t i o n of ferritin as compared w i t h the tissues fixed s u b s e q u e n t to t r e a t m e n t w i t h the con] agate. T r e a t m e n t on Copper Grids. Bacterial, viral, and m y c o t i c agents were fixed in 10% formalin and drops placed on f o r m v a r and carbon covered grids 4. The grids were air-dried, specific i m m u n e c o n j u g a t e applied, t h e n washed several times in deionized water. Non-specific Reactions. Horse spleen ferritin ~ was diluted w i t h p h o s p h a t e buffered saline, p H 7.2, to c o n t a i n 10 m g / m I of protein and c o n j u g a t e d w i t h fluorescein isot h i o c y a n a t e 8. 0.5 ml of the ferritin fluorescein c o n j u g a t e was injected i n t r a p e r i t o n e a l l y into m a t u r e mice. Sections of liver, e m b e d d e d in paraffin, revealed t h a t the ferritinfluorescein c o n j u g a t e was p h a g o c y t i z e d b y the K u p f f e r ceils non-specifically (i.e. in regard to i m m u n e reactions). O b s e r v a t i o n s were m a d e using an u l t r a v i o l e t light source and filters p r e v i o u s l y outlined 6. Discussion. The proper use of f i x a t i v e s is of great imp o r t a n c e in t h e d e t e c t i o n of a n t i g e n s b y i m m u n e electron microscopy. P e n e t r a t i o n of a n t i b o d y i n t o cellular m a t e r i a l is d e p e n d e n t upon a d e q u a t e fixation, Correlation can be

the b e h a v i o u r of the D N A - p l a s m of the bacterial nucleus. I t is found t h a t those conditions which produce coarse coagulation of the free D N A also produce electron o p a q u e bodies of v a r i a b l e aspect inside t h e bacterial nucleus. The c o a g u l a t i o n figures o b t a i n e d in b o t h situations are v e r y similar. This is a further direct evidence in f a v o u r of the v i e w t h a t t h e bacterial nucleus is an assembly of fine D N A - c o n t a i n i n g fibrilla in a highly h y d r a t e d plasma. ~V. These obs e r v a t i o n s are consistent w i t h those of HIRAMOTO et al. 7. F i x a t i o n prior to t r e a t m e n t w i t h the ferritin c o n j u g a t e p r e v e n t s pinocytosis, resulting in a m a r k e d d i m i n u t i o n of the u n r e a c t e d c o n j u g a t e as c o m p a r e d w i t h m a t e r i a l fixed after t r e a t m e n t w i t h t h e conjugate. F o r m a l i n generally fulfills t h e criteria as an excellent f i x a t i v e for this technique. I t does n o t t e n d to alter the antigenic c o m p o n e n t s , is germicidal for m o s t pathogens, and preserves t h e cellular architecture. F i x a t i v e s generally used in electron m i c r o s c o p y are p o t e n t oxidants, and t h o r o u g h screening should be m a d e to d e t e r m i n e their effect on the antigenic s y s t e m to be used. This does n o t preclude the use of these o x i d a n t s to enhance the a r c h i t e c t u r e if applied s u b s e q u e n t to the i m m u n e reaction. I n t h e d e t e c t i o n of sites of toxin a c t i v i t y and in rive experiments, the use of a d e q u a t e controls c a n n o t be t o o g r e a t l y stressed. F e r r i t i n can be observed u n d e r the electron microscope in n o r m a l cellular m a t e r i a l (HAMPTON 8, KUFF a n d DALTON 9, and BESSIS and BRETONGORIUS t°) as well as ferritin c o n j u g a t e which has been non-specifically p h a g o c y t i z e d b y t h e r e t i c u l e - e n d o t h e l i a l system. Therefore, in correlation w i t h n o n i m m u n e conj u g a t e controls, h i s t o c h e m i c a l methods, fiuoresceinlabeled ferritin and electron m i c r o s c o p y should be utilized. These will n o t o n l y p r o v i d e a control of t h e i m m u n e
